Antimutagenic activity of cashew apple (Anacardium occidentale Sapindales, Anacardiaceae) fresh juice and processed juice (cajuína) against methyl methanesulfonate, 4-nitroquinoline N-oxide and benzo[a]pyrene Genet. Mol. Biol.
Melo-Cavalcante,Ana Amelia; Picada,Jaqueline N.; Rubensam,Gabriel; Henriques,João A.P..
Cashew apple juice (CAJ), produced from the native Brazilian cashew tree (Anacardium occidentale), and has been reported to have antibacterial, antifungal, antitumor, antioxidant and antimutagenic properties. Both the fresh unprocessed juice and the processed juice (cajuína in Portuguese) has been shown to consist of a complex mixture containing high concentrations of anacardic and ascorbic acids plus several carotenoids, phenolic compounds and metals. We assessed both types of juice for their antimutagenic properties against the direct mutagens methyl methanesulfonate (MMS) and 4-nitroquinoline-N-oxide (4-NQO) and the indirect mutagen benzo[a]pyrene (BaP) using pre-treatment, co-treatment and post-treatment assays with Salmonella typhimurium strains...

Induction and elimination of bulky benzo[a]pyrene-related DNA adducts and 8-oxodGuo in mussels Mytilus galloprovincialis exposed in vivo to B[a]P-contaminated feed ArchiMer
Akcha, Farida; Burgeot, Thierry; Budzinski, H; Pfohla Leszkowicz, A; Narbonne, J.
With the aim of developing biomarkers of polycyclic aromatic hydrocarbon (PAH) genotoxicity for biomonitoring of the marine environment, the formation of DNA adducts and oxidative DNA damage was studied in the sentinel organism Mytilus. Mussels M. galloprovincialis were exposed for 21 d to different doses of benzo[a]pyrene (B[a]P) (50 and 100 mg B[a]P kg(-1) dry wt mussel d(-1)) via trophic intake. The kinetics of B[a]P concentration in whole mussel tissues was determined in both groups, and a dose-response study was conducted for both gill B[a]P-related DNA adducts and 8-oxo-7, 8-dihydro-2'-deoxyguanosine (8-oxodGuo) formation in gill and digestive gland DNA. Exposure was followed by a 10 d depuration period to study mussel recovery. Although B[a]P was...

Investigating the relationship between embryotoxic and genotoxic effects of benzo[alpha]pyrene, 17 alpha-ethinylestradiol and endosulfan on Crassostrea gigas embryos ArchiMer
Wessel, Nathalie; Rousseau, Sabrina; Caisey, Xavier; Quiniou, Francoise; Akcha, Farida.
Genotoxicity biomarkers are widely measured in ecotoxicology as molecular toxic endpoints of major environmental pollutants. However, the long-term consequences of such damage still have to be elucidated. Some authors have suggested that the accumulation of unrepaired DNA lesions could explain the embryotoxicity of certain chemical pollutants. As embryotoxicity exerts a direct impact on the recruitment rate, genotoxicity could be closely related to disturbances of ecological concern and produce a possible impact upon population dynamics. The aim of the present work was to study the genotoxicity and the embryotoxicity of three relevant pollutants for oyster embryos: the polycyclic aromatic hydrocarbon, benzo[a]pyrene (BaP), the synthetic estrogenic...

The toxicity of benzo[a]pyrene on sole (Solea solea) hepatocytes: assessment of genotoxic and enzymatic effects ArchiMer
Wessel, Nathalie; Menard, Dominique; Pichavant-rafini, K.; Ollivier, H.; Le Goff, Jean-michel; Burgeot, Thierry; Akcha, Farida.
The benzo[a]pyrene is a polycyclic aromatic hydrocarbon known to be genotoxic, mutagenic and carcinogenic in higher vertebrates. The aim of this study was to evaluate in vitro the enzymatic and genotoxic effects of BaP in a benthic fish species, Solea solea. Sole hepatocytes were exposed to BaP in order to measure the modulation of ethoxyresorufin-o-deethylase (EROD) activity and the DNA strand breaks induced by BaP metabolism. Exposures were performed in both culture flasks and microplate wells in order to check for the possible miniaturization of the exposure system. Moreover, sole liver microsomes were exposed to BaP in the presence of standard DNA in order to assess the potential formation of DNA adducts in sole. The results demonstrated the ability of...
